筋萎縮性側索硬化症 以下 ALS は運動ニューロンを選択的に侵す、原因不明かつ治療法のない神経変性疾患です。 そこで、当社が開発した脳と脊髄の広範な領域の神経細胞に遺伝子導入可能な改変型アデノ随伴ウイルス AAV ベクターを使用してヒトADAR2を脳と脊髄の運動ニューロンに送達し、低下したADAR2活性を正常化させることにより、運動ニューロン死 変性と脱落 を阻止し、症状の進行を食い止めます。 孤発性筋萎縮性側索硬化症 ALS では、運動ニューロンでRNA編集酵素であるadenosine deaminase acting on RNA2 ADAR2 の発現が低下し、AMPA受容体 グルタミン酸受容体サブタイプ のGluA2サブユニットに本来起こるべきRNA編集を欠いた未編集型GluA2が発現しています。 当社の遺伝子治療では、治療遺伝子を神経細胞に導入するベクターとしてアデノ随伴ウイルスadeno-associated virus (AAV) を使用します。 この反応により遺伝子にコードされたグルタミン Q コドン CAG がCIGに変換されます。
