Tuesday, August 29, 2006. Week 10 - DAPHNE- Gel Extraction and Column purification. 1 Excise the DNA band of interest by viewing under UV and place them into individually labelled microcentrifuge tubes. 2Add 300ul of buffer GB to each tube. 3 Incubate for 10 min at 55 degC with occasional mixing by vortexing. this is to help dissolve the agarose gel. 4 Transfer the mixture to a spin column, centrifuge for 2 min, discard the flow-thru. 7Apply 30ul of water, let stand for 5-10 min to elute out the DNA.
